Fernando Meirelles wanted the cast to consist of people who had never acted before and professional actors who were not widely known. One such actor was Matheus Nachtergaele, whom Meirelles had seen in a play. While Meirelles was working on the script, however, Nachtergaele suddenly became a huge star in Brazil after starring in the hit film A Dog's Will (2000). Meirelles was a little disappointed at first but Nachtergaele promised him that he would disappear into the role of Sandro Cenoura so completely that his stardom would not distract from the film. He actually moved to the real Cidade de Deus where most of the cast lived and lived there for three months to prepare for the role.

Originally, the idea for the Arquillians was meant for a minor character, a bartender named Chuckie. To prove that he was an alien, he was to shoot a beam of light from his head. Rick Baker suggested that he was a tiny alien living inside the head of a robot body. Ultimately the character was cut, but the producers liked Baker's idea so much, they decided to use it for characters that were more important to the plot.

Composer Elmer Bernstein was hired due to an enthusiastic recommendation by the film's Editor Malcolm Campbell to Edward Norton, with whom Campbell had worked with when he was the primary editor for John Landis' films and Bernstein had scored for a period of ten or more years. Those films include An American Werewolf In London, Trading Places, Michael Jackson's Thriller, Spies Like Us, and Three Amigos! Campbell was an assistant editor when Bernstein first collaborated with Landis on National Lampoon's Animal House and also on Blues Brothers, where Bernstein provided the very brief "God Music" for the film. Bernstein was not involved with Twilight Zone The Movie which Campbell edited Landis' segment since the entire film was scored by the late Jerry Goldsmith.
